Utjecaj temperaturnog stresa na klorofilnu fluorescenciju kod krumpira

Abstract

Krumpir je jedna od najvažnijih prehrambenih kultura za većinu svjetske populacije, a ujedno je vrlo osjetljiv na abiotske stresove. Kao posljedica klimatskih promjena očekuje se znatno povećanje temperature u narednim desetljećima čime se povećava rizik od stresa visokih temperatura, jednog od najznačajnijih abiotskih stresova. Cilj ovog rada je utvrditi utjecaj visokih temperatura na parametre klorofilne fluorescencije kod krumpira. Klorofilna fluorescencija je jedna od najkorištenijih neinvazivnih metoda procijene stresa kod biljaka. U pokusu je korištena sorta krumpira Bella Rosa. Nakon klijanja gomolja odvojeno je po sedam biljaka za svaki od četiri temperaturna tretmana; 20/15 °C, 25/20 °C, 30/25°C i 35/30 °C. Nakon deset dana uzgoja biljaka pri svakom tretmanu određivana je klorofilna fluorescencija. Mjerenja su obavljana slikanjem biljaka pomoću CropReporterTM (PhenoVation B.V., Wageningen, The Netherlands) uređaja. Podaci su analizirani jednosmjernom analizom varijance (ONE WAY ANOVA) u statističkom programu SAS. Stres visokih temperatura negativno je utjecao na većinu mjerenih parametara klorofilne fluorescencije osim na maksimalnu efikasnost PSII (Fv/Fm), najjače je utjecao na vrijednosti efektivne efikasnosti PSII (Fq'/Fm') te vrijednosti nefotokemijskog čišćenja (NPQ). Pri čemu je vrijednost Fq'/Fm' padala s porastom temperature a vrijednost NPQ rasla s porastom temperature. Ovi rezultati upućuju na zaključak da je smanjena efikasnost korištenja svjetlosne energije rezultat zatvaranja reakcijskih centara fotosustava pri visokim temperaturama te stoga povećanja nefotokemijsokg čišćenja.

Abstract (english)

Potato is one of the most important food crops for most of the world’s population and is highly vulnerable to abiotic stresses. As a result of climate change, a significant increase in temperature is expected in the coming decades, increasing the risk of high temperature stress, one of the most significant abiotic stresses. The aim of this study was to determine the influence of high temperatures on the parameters of chlorophyll fluorescence in potatoes. Chlorophyll fluorescence is one of the most widely used non-invasive methods for the assessment of the plant stress. For the purpose of the experiment, tubers of the Bella Rosa variety were planted. After germination, seven plants were selected for further grow in one of the four temperature treatments: 20/15°C, 25/20°C, 30/25°C and 35/30°C. After ten days, chlorophyll fluorescence parameters were determined. Measurements were performed by imaging the plants using a CropReporterTM (PhenoVation B.V., Wageningen, The Netherlands) device. Data were analyzed by one-way analysis of variance (ONE WAY ANOVA) in the SAS statistical program. High temperature stress negatively affected most of the measured parameters of chlorophyll fluorescence except for the maximum efficiency of PSII (Fv/Fm). Most affected were the values of the effective efficiency of PSII (Fq'/Fm') and the values of nonphotochemical quenching (NPQ). Where the value of Fq'/Fm' decreased with increasing temperature and the value of NPQ increased with increasing temperature. These results suggest that under the high temperatures potato has lower light use efficiency due to closure of PSII reaction centers and excess light energy is dissipated through the nonphotochemical quenching.
