Abstract

U Republici Hrvatskoj zabilježen je visok udio zaraženih nasada autohtonih sorata vinove loze pri čemu zaraze virusima predstavljaju najveći problem. Pomoću in vitro metoda provodi se postupak oslobađanja vinove loze od virusa. Cilj istraživanja bio je utvrditi utjecaj sanitarnog statusa kod klonova predmetnih sorata na rast i razvoj eksplanatata. Klonovi sorata ˈPlavac maliˈ i ˈPošipˈ različitog sanitarnog statusa s obzirom na zaraženost virusima (GVA, GFLV, GLRaV-1, GLRaV-1+GLRaV-3) uvedeni su u kulturu tkiva korištenjem različite kategorije pupova sa zeljastih mladica (a - apikalni meristem, b - 2. i 3. pup na početnom izdanku i c - 4. i 5. pup na početnom izdanku) te su pomoću metode kulture meristema inokulirani na medij (MS+0,5 mg/L BAP + 0.05 mg/l IAA). Rast i razvoj eksplantata u kulturi tkiva mjeren je nakon četiri i osam tjedana kultivacije. Rezultati pokazuju signifikantan utjecaj sorte u rastu i razvoju eksplantata in vitro, dok sanitarni status klonova unutar sorata te kategorija pupova nisu signifikantno utjecali na mjerene parametre visine, broja nodija i pojave kalusa, osim što je kategorija pupa signifikantno utjecala na stvaranje kalusnog tkiva. Dobivene rezultate potrebno je nadopuniti sa daljnjim istraživanjima ovdje testiranih sorata.

Abstract (english)

In the Republic of Croatia, a high percentage of infected vineyards of autochthonous grapevine varieties has been recorded, with virus infections representing the biggest problem. Using in vitro methods, the process of freeing the vine from the virus is carried out. With this research, the aim was to determine the impact of the sanitary status of clones of varieties "Plavac mali" and "Pošip" on the growth and development of explants. Clones of the varieties "Plavac mali" and "Pošip" with different sanitary statuses in terms of virus infections (GVA, GFLV, GLRaV-1, GLRaV-1+GLRaV-3) were introduced into tissue culture using different categories of buds from the initial shoots (a - apical meristem, b - 2nd and 3rd bud on the initial shoot, and c - 4th and 5th bud on the initial shoot), and they were inoculated onto a medium (MS+0.5 mg/L BAP + 0.05 mg/l IAA) using the meristem culture method. The growth and development of explants in tissue culture were measured after four and eight weeks of cultivation. The results show a significant impact of the variety on the in vitro growth and development of explants, while the sanitary status of clones within the varieties and the categories of buds did not significantly affect the measured parameters of height, number of nodes, and the occurrence of calluses, except that the bud category significantly influenced the formation of callus tissue. Further research is needed to complement the obtained results regarding the tested varieties.
