Abstract

Biljka kapara zbog niske klijavosti i visokog stupnja heterozigotnosti u sjemenu neprikladna je za razmnožavanje konvencionalnim metodama. Kao jedno od rješenja koje bi pospješilo i ubrzalo razmnožavanje ove biljne vrste je mikropropagacija. Najčešće korišteni citokinin u svrhu mikropropagacije kapara (C. orientalis Veill.) je 6-benziloaminopurin (BAP). Metatopolin (mT) se smatra alternativom BAP-u jer osigurava visoke stope umnožavanja uz manje negativnih posljedica. Upotreba citokinina koja je nužna za multiplikaciju in vitro može dovesti do stresnih uvjeta i povećane proizvodnje reaktivnih kisikovih oblika (ROS) koji ograničavaju mikropropagaciju jer dovode do morfofizioloških promjena i aktivacije enzima stresa. Cilj rada je bio odrediti učinak vrste i doze citokinina na stopu multiplikacije, lisnu površinu i razinu aktivnosti enzima stresa i ukupnih proteina u izdancima kapara u in vitro uvjetima.

Abstract (english)

Due to low germination and high degree of heterozygosity in the seeds, the caper plant is not suitable for propagation by conventional methods. Micropropagation is one of the solutions that would promote and speed up the reproduction of this plant species.The most used cytokinin for the purpose of micropropagation of caper (C. orientalis Veill.) is 6-benzylaminopurine (BAP). Metatopolin (mT) is considered as an alternative to BAP because it provides high multiplication rates with fewer negative consequences. The use of cytokinin, which is necessary for multiplication in vitro, can lead to stressful conditions and increased production of reactive oxygen species (ROS) that limit micropropagation. They lead to morphophysiological changes and activation of stress enzymes. The aim of the work was to determine the effect of the type and dose of cytokinin on the multiplication rate, leaf area and level of stress enzyme and protein activity in caper shoots in vitro.
